Genome-Wide Analysis of DNA Cytosine Methylation in Two Poplars

DNA methylation is an important epigenetic modification that has been implicated in many biological processes. Methylation changes during stress responses and developmental processes have been well studied. Here, we present a novel comparison of methylation patterns in two poplar species. We detected the global methylation patterns in leaves of Populus trichocarpa and Populus euphratica by methylated DNA immune precipitation sequencing (MeDIPseq). A total of eleven significant modules were detected in the two species. Our results indicate that the methylation of CG/CHH/CHG contexts was much more significant in P. trichocarpa than in P. euphratica, but methylation changes in response to saline stress were consistent in both species.

During their life cycles, plants are constantly challenged by biological and abiotic environmental disturbances. In response to these disturbances, plants have developed genome-wide changes in gene expression to regulate their physiological and developmental mechanisms. Recent studies have suggested that DNA methylation plays a vital role in plant stress responses. Environmental stimulation, including salt and water stress, can lead to demethylation of the coding regions of certain genes, subsequently activating their expression. In this study, we applied MeDIP-seq to profile the DNA methylation status of P. trichocarpa and P. euphratica under salt stress and control treatments. Our MeDIP-seq data indicate that DNA methylation is more common in gene body regions than in other gene regions, consistent with previous results. A total of 157.8 million reads were generated, 132.8 million of which could be mapped to the reference genome. You can read the article on our journal website.
